Identification of potential target genes of USP22 via ChIP-seq and RNA-seq analysis in HeLa cells Genet. Mol. Biol.

Abstract The ubiquitin-specific protease 22 (USP22) is an oncogene and its expression is upregulated in many types of cancer. In the nucleus, USP22 functions as one subunit of the SAGA to regulate gene transcription. However, the genome-wide USP22 binding sites and its direct target genes are yet clear. In this study, we characterized the potential genomic binding sites of UPS22 and GCN5 by ChIP-seq using specific antibodies in HeLa cells. There were 408 overlapping putative target genes bound by both USP22 and GCN5. Motif analysis showed that the sequences bound by USP22 and GCN5 shared two common motifs. Gene ontology (GO) and pathway analysis indicated that the genes targeted by USP22 and GCN5 were involved in different physiological processes and...

Propranolol inhibits the proliferation, migration and tube formation of hemangioma cells through HIF-1α dependent mechanisms BJMBR
Chen,Y.Z.; Bai,N.; Bi,J.H.; Liu,X.W.; Xu,G.Q.; Zhang,L.F.; Li,X.Q.; Huo,R..
The aim of this study was to investigate the mechanism of propranolol on the regression of hemangiomas. Propranolol-treated hemangioma tissues were collected and the expression of hypoxia inducible factor-1α (HIF-1α) was examined. We also established HIF-1α overexpression and knockdown hemangioma cells, and determined the effects of HIF-1α on the hemangioma cells proliferation, apoptosis, migration and tube formation. Significantly increased HIF-1α level was found in the hemangioma tissues compared to that in normal vascular tissues, whereas propranolol treatment decreased the HIF-1α level in hemangioma tissues in a time- and dose-dependent manner. Moreover, propranolol treatment significantly decreased cell proliferation, migration and tube formation as...

The RNA interference revolution BJMBR
Lenz,G..
The discovery of double-stranded RNA-mediated gene silencing has rapidly led to its use as a method of choice for blocking a gene, and has turned it into one of the most discussed topics in cell biology. Although still in its infancy, the field of RNA interference has already produced a vast array of results, mainly in Caenorhabditis elegans, but recently also in mammalian systems. Micro-RNAs are short hairpins of RNA capable of blocking translation, which are transcribed from genomic DNA and are implicated in several aspects from development to cell signaling. The present review discusses the main methods used for gene silencing in cell culture and animal models, including the selection of target sequences, delivery methods and strategies for a successful...

Toxicity of Field-Aged Permethrin-Loaded Attracticides on Choristoneura rosaceana (HARRIS) and Pandemis pyrusana Kearfott (Lepidoptera: Tortricidae) Adult Males and Females Chilean J. Agric. Res.
Curkovic,Tomislav; Brunner,Jay F; J. Brown,John.
Lethal and sub lethal effects were evaluated in the laboratory on Choristoneura rosaceana (Harris) and Pandemis pyrusana Kearfott males exposed to an attracticide loaded with permethrin and aged in the field. The effect of pairing conspecific females with previously intoxicated males was also evaluated. In both species a significantly greater mortality of male moths (≥ 95%) was observed 24 h after exposure to t he attracticide formulation. Likewise, high levels of knock down (≥ 75%) were observed in males of both species 1 h after attracticide exposure. In females, knock down was ≥ 2.5% and mortality ≥ 3.3% after pairing with intoxicated males. Leg autotomy in males was at least 73% in C. rosaceana and ≥...
